Now that Dune's Significance has been, confirmed, it appears that the Director Denis Villeneuve will not waste any time getting the ball rolling on a follow up.

The installment earned $400, million at the world- wide box office and picked up to 10 Oscar nomination

Dune: Part Two is set to complete Denis Villeneuve's adaptation of the original book, that does not necessarily mean the film franchise will end as the director of Dune has already taken much interest in tackling the Sequel Novel.

The MoonKnight actor, Oscar Issac as Duke Leto and Sharon Duncan-Brewster's , Dr. Lier -Kynes will probably not return as they passed away in Dune:Part One.
However, if Dune:Part Two also adapts part of Dune's second book, Dune MessiahJason Momoa as Duncan Idaho could return as a GholaClone.

Though, it's probably a good bet the rebooted Duncan (also known as "Hayt") will not appear until the third film which is Dune:Part Three  that is in the Trilogy.

Dune:Part Two Plot.

Dune:Part One   adapted around the first 500 pages of the book.
Dune: Part Two is suppose to cover the rest of Dune by Frank Herbert.
If the book follows closely, Dune:Part Two will continue telling Paul's story, Chronicling his time with the Firemen, his relationships with Chani and Princess Irulan, and his plans to take down the empire.
Denis Villeneuve also said the scene where Josh Brolin's Gurney Halleck sings was cut from the first movie. The director said he hopes to find a place for it in Dune:Part Two.

I will not like to spoil  this movie, I just want to give you a hint on how the movie is going to be like....

Will There Be A Dune: Part Three.
While Dune:Part One &Part Two will cover the first of Frank Herbert's series, there could be a third movie based on the second book, Dune Messiah.

The Interview that was held between Denis Villeneuve & Inverse hinted at a third movie, and if both Part One &Two perform well, it's not Far-fetched that Warner Bros. would order a third film to round out of Trilogy.
